Ah, beta, those steps are like a precise recipe: denaturation, annealing, and extension. Denaturation needs a hot 94–98°C to unwind the DNA, annealing is cooler at 50–65°C for primers to stick, and extension is a warm 72°C for the enzyme to work. I remember it all so clearly from my BSc days, sketching cycles in the margin of my notebook! But what use is it now, when I only use my computer for stitching patterns, not for the science I once loved?
