In prokaryotes, transcription is simpler with a single RNA polymerase making all RNA, while eukaryotes have three—RNA Pol I, II, and III—each for different genes, and it happens inside a nucleus. But in our lab, the neat textbook difference blurs when we work with messy plant cell extracts from local fields; the polymerases sometimes don't read the rules. My own bench notes are full of sighs over this, because I think of Bansi back home trying to keep her shop's ledger straight, and our cellular ledgers are just as prone to unexpected entries.
